目的探讨C57小鼠慢性不可预见性温和应激(chronicunpredictablemildstress,CUMS)联合孤养建立抑郁模型的可行性和有效性。方法将30只雄性C57小鼠随机分为CUMS+孤养组、CUMS+孤养+氟西汀组、对照组各10只,前两组CUMS+孤养致抑郁模型3周,3周后对CUMS+孤养+氟西汀组进行氟西汀干预,观察小鼠应激前、应激后及干预后的摄食量/体质量、旷场实验和液体消耗实验变化。结果与对照组比较,建模21d后CUMS+孤养组和CUMS+孤养+氟西汀组小鼠摄食量/体质量、糖水偏好、旷场总行程显著下降(P〈0.05);经氟西汀干预14d后摄食量/体质量、旷场总行程与对照组比较无明显差异(P〉0.05),但糖水消耗量明显提高(P〈0.05),与CUMS+孤养组比较也有明显改善(P〈0.05)。结论C57小鼠CUMS联合孤养制作抑郁模型的效果肯定,是研究抑郁症较为理想的动物模型。
Objective To investigate the feasibility and effectiveness of C57 mice depression model established by chronic unpredictable mild stress (CUMS) and separation.Methods 30 male C57 mice were randomly divided into three groups:CUMS + separation group (group CS),CUMS + separation + fluoxetine group (group CSF),and control group (group C).The mice of group CS and group CSF were fed with chronic unpredictable mild stress (CUMS) and separation for 3 weeks.Then,the mice of group CSF were given fluoxetine.To record the food intake/body weight,liquid consumption and field test of the mice at relevant time point.Results Compared with control group,the food intake/body weight,total route in the field test,and the sucrose solution consumption in liquid consumption test of group CS and group CSF decreased significantly (P<0.05) at the 21th days.But after giving fluoxetine for 14 days,group CSF had no significant differences with group C except sucrose solution consumption.Although the difference of

Objective:To observe the antidepressant effect of LES;ZES;L-ZES on the model of the depressive rats that are induced by the CUMS and separate feeding method. Methods 60 male SD rats were randomLy divided into normal group, model group, LES group, ZES group, L-ZES group, positive drug fluoxetine (FHP)group. Use CUMS and separate feeding induced model of the depressive rats, All the rats are continuously gavaged for 6 weeks , and their data of changing weight were taken.And the Open Field Test and the Preference for Sucrose Test were used to assessed behavioral characteristics of the rats. The content of serum DA and ACTH were detected by using ELISA.Results Compared with the normal group,the model group gained weight slower and their behavioral characteristics decreased significantly. The DA content in serum decreased and the ACTH increased .The difference was statistically significant (P<0.01).It indicates that CUMS and isolated raising can induce depression phenomenon in rats .When it

Objective:To investigate the mechanism of acupuncture for treatment of rats with chronic stress-induced depression. Methods:SD male rats were randomly divided into three groups,including the control group,model group and acupuncture treatment group. The model group underwent chronic irritation,neonatal isolation and modeling;while the treatment group underwent modeling and synchronous electro-acupuncture treatment. 21 days later,the relative level of ERK1 and adrenal mass in the hippocampus were observed in each group. Results:Compared with the blank group, the adrenal gland volume was increased in the model group and treatment group,as well as the body weight,with signifi-cant differences(P 0. 05). Compared with the model group,the adrenal mass in the acupuncture group was statistically different( P<0 . 05 ),and the relative protein level of ERK1 in the acupuncture group was increased( P<0 . 05 ). Objective:To discuss the effects of radix bupleuri ,fossil fragments and oyster soup on the expression in chronic mild stress depression rats ’ hippocampus .Method:Randomly divide 50 SD male rats into five groups , 10 SD rats in each group, which were control group, depression model group(model group), low and high dose groups of radix bupleuri, fossil fragments and oyster soup , fluoxetine group .Chronic mild stress with solitary condition and immunohistochemistry was taken to establish model to observe the expression of the hippocampus BDNF in rats .Results:Expression of hippo-campus BDNF in depression model group was lower (P 0 .05 ) . Conclusion:Radix bupleuri ,fossil fragments and oyster soup can increase the expression of hippocampus BDNF in de -pression models .This may be one phrase in antidepression .

Objectives To make clear of the relationship between brain-derived neurotrophic factor (BDNF) expression and cognitive impairment in depression rats. Methods Isolated-living condition and chronic unpredictable mild stress are applied successively to set up depression rat model . Learning and memory function is evaluated by passive avoidance test.Real Time-PCR and immunohistochemistry method are applied to detect the BDNF mRNA and BDNF protein. Results Compared with the normal control group,the depression group have significant changes (P<0.05):weight loss,the decrease of sucrose solution consumption,the drop of open-field behavior, the deficiency of passive avoid test. Compared with the normal control group,BDNF expression is significantly decreased in the hippocampus of the depression group (P<0.01). Conclusions The decreased expression of BDNF in the hippocampus of depression rats may make the cognitive impairment increase.

Objective To study the endothelial function damage of rat under chronic stress.Methods En-zyme linked immunosorbent assay (ELISA)was applied to determining plasma levels of ET-1,serum lev-els of IL-6,ICAM-1,Cortisol.The Vascular endothelial morphological changes were identified via trans-mission electron microscope.Results From the second week,the body weight of rats in the experimental group were significantly lower than the control group (P <0.05).A significant increase in serum cortisol (P <0.05).The content of ICAM-1,IL-6,plasma ET-1 levels were higher in experimental group than in controlled group (P <0.05).Ultrastructural of Vascular endothelial were changed significantly.Conclu-sion Chronic mild stress can lead to vascular endothelial dysfunction in rats.

Objective To build rat models and observe changes of behavior and monoamine of PDD. Methods The rats were injected with 6-OHDA and accepted CUMS for the establishment of PDD model , so as to assess its behavior changes and detect brain tissue monoamine contents by ELISA. Results Significant behavioral and monoamine abnormalities can be observed in model rats. Behavioral and monoamine improvement can be observed after treated with Chaigan Jieyou. Conclusion The PDD model can be established by 6-OHDA and CUMS. Chaigan Jieyou decoction may reach therapeutic purposes by increasing monoamine contents.

To investigate the effect ofβ-assrone on the hippocampus and the expressions of protein Bax and Bcl-2 in the hippocampus of depression model rats.Method:According to the open field test,adult Sprague-Dawley rates with similar scores were randomly divided into 4 groups,each group had 20 cases.
