目的为皮上划痕人用布氏菌活疫苗筛选存活率高、无明胶冻干稳定剂。方法以冻干活菌存活率为指标,对甘油、甘露醇、蔗糖、葡萄糖、乳糖、谷氨酸钠、甘氨酸、谷氨酸、脯氨酸和硫脲等10种稳定剂通过单因素筛选法,筛选出冻干存活率高的4种单因素稳定剂成分;将4种单因素稳定剂成分进行正交试验优化,筛选出最优稳定剂组合。结果单因素试验结果显示,甘油、葡萄糖、谷氨酸钠和硫脲4种稳定剂成分冻干后活菌存活率较高,对布氏菌活疫苗具有良好保护效果。通过正交试验筛选出最优稳定剂配方中四组分的质量分数分别为甘油1.5%、葡萄糖5%、硫脲1.5%、谷氨酸钠1.0%,该配方的冻干存活率可达81.5%。结论无明胶冻干稳定剂对布氏菌活疫苗具有较好的保护作用。
Objective To screen a gelatin-free protective agent for freeze-dried live Brucella vaccine. Methods The pro-tective effects of 10 different protective agents were compared with single factor such as glycerol, mannitol, sucrose, glu-cose, lactose, glutamine sodium, glycine, glutamic acid, proline and thiourea, and the orthogonal design was carried out. The viable count as a index was detected for selecting a better formula. Results The tested results showed that glycerol, glucose, glutamic sodium and thiourea is the good protective agent. The formula containing 1. 5% glycerol, 5% glucose, 1. 5% thiourea and 1. 0% glutamic sodium is the most optimal formula. The viable rate of freeeze-dried vaccine after adding this formula agent could be achived 81. 5%. Conclusions The gelatin-free freeze-dried cryoprotectant is of the good protective effect on the live Brucella vaccine.

Using single factor and orthogonal test methods, we studied the stabilizing effects of different stabilizers and their compound stabilizer on the cloudy juice of yacon. The results showed that, the stabilizing effects of the single stabilizer from high to low were sodium alginate, pectin and gelatin, the stabilizing effect of the compound of the three kind of stabilizers was better than that of single stabilizer, the stabilizing effect of compound stabilizer 0.05%sodium alginate+0.05%pectin+0.04%gelatin was the best, and the naturally layered rate was 0.70%when stored at 4℃for 40 d.

Objective To investigate the effets of three mood stabilizers on ouabain?induced ERK1/2 phosphorylation in astrocytes. Methods As?trocytes were treated with different agents and divided into different groups accordingly,namely,the control group with saline,the group with oua?bain,the group with mood stabilizers(lithium carbonate,carbamazepine,sodium valproate)and the group with ouabain+mood stabilizers. The phosphorylation of ERK1/2 in each group was analyzed by Western blot. Results Compared with saline and mood stabilizer groups,the phosphoryla?tion of ERK1/2 was increased in the ouabain group,with statistical significance(P<0.05). There was no significant difference in ERK1/2 phosphoryla?tion between the group with ouabain+mood stabilizers and the control or mood stabilizer group. Conclusion The three kinds of mood stabilizers can inhibit ouabain?induced ERK1/2 phosphorylation in astrocytes.

The influence of two kinds of homemade Ca/Zn composite thermal stabilizers with high efficiency on the thermal stabilities and processing rheological properties of poly (vinyl chloride)(PVC) was evaluated by means of congo red test,thermal aging oven test,rheological test. The results show that the thermal stabilities and rheological properties of PVC with calcium/zinc 2,2-bis(hydroxymethyl) propionate composite thermal stabilizer is better than the products sold in the market,there is no noticeable discoloration for 80 min at 180℃,the congo red test paper do not discolor until 69.5 minutes later,the various parameters of the rheological properties are good. The properties of the other homemade stabilizer using Ca/Zn stearate is comparable with the foreign product.

Effect of Mg(OH)2 on rheological properties and processing stability of poly(vinyl chloride)(PVC) was discussed, in which Ca-Zn-hydrotalcite composite thermal stabilizer or composite thermal stabilizer of lead sulfate tribasic and lead phosphate dibasic,acrylate copolymer(ACR) KM 310,ACR 401,PE wax and stearic acid were used as auxiliary. When content of thermal stabilizer,ACR KM 310,ACR 401,PE wax and stearic acid were kept constant,the minimum torques,maximum torques,the balance torque and the processing energy consumption increase,but the processing thermal stabilities reduce with increasing of Mg(OH)2 content. The maximum torque is lower than 38.3 N · m and thermal stabilization time exceed 20 min,in which Mg(OH)2 content is below 10 phr. While Mg(OH)2 content exceeds 20 phr,the maximum torque is more than 40 N · m and thermal stabilization time is lower than 14 min. The insoluble sulfur was prepared , and the effects of the type and dosage of stabilizers , re-action temperature and thermostatic reaction time on content and high-temperature stability of the insolu-ble sulfur were studied.The obtained product was characterized by scanning electron microscope , X-ray diffraction and differential scanning calorime-try.The results showed that the efficiency of antioxi-dant 264/calcium stearate complex stabilizer was better than that of antioxidant 264 or calcium stea-rate single stabilizer.Under the optimized condition of stabilizer/sulfur ( mass ratio ) 1/1 000 , reaction temperature 240 ℃ and thermostatic reaction time 30 min, the mass fraction and high-temperature sta-bility of the insoluble sulfur reached up to 46.24%and 60.24%, respectively.
