采用超声波辅助萃取法提取,高效液相色谱法测定了锁阳(Cynomorium songaricum)中的熊果酸含量。在单因素试验基础上,以乙醇体积分数、料液比、超声时间、超声功率为研究对象,进行了4因素3水平正交试验。结果表明,熊果酸在4.0~16.0μg/μL范围内呈良好线性关系,回归方程为y=622.7x-85.517,r=0.999 8,加样回收率为98.08%。锁阳中熊果酸最佳提取工艺为:超声功率150 W,料液比1∶25,85%(V/V,下同)乙醇超声提取30 min,在此条件下,锁阳中熊果酸含量为1.53%。该工艺能有效提取锁阳中熊果酸,为评价中药材中的熊果酸水平提供参考。
Ursolic acid in Cynomorium songaricum was ultrasonically extracted and detected by HPLC. According to singlefactor experiments,three levels of four factors including ethanol concentration,material -liquid ratio,ultrasonic time, and ultrasonic power were selected for the orthogonal design.The results showed that the calibration curve of ursolic acid was linear (r=0.999 8) in the range of 4.0~16.0 μg/μL.The equation was y=622.7x-85.517,r=0.999 8,and recovery was 98.08%. The optimum extraction was carried out in 85%(V / V) ethanol for 30 min with the ultrasonic power of 150 W,and the material-liquid ratio 1:25.Under the optimal conditions,the content of ursolic acid was 1.53%.The method can extract ursolic acid efficiently,and provide a reference for evaluation ursolic acid in medicinal materials.

Based on the PEG experiment for seeds and pot without water experiment for the seedling , we designed the drought stress simulation experiments of Helianthemum songaricum seeds and seedlings , and studied the effects of drought stress on seed germination and seedling physiological characteristics of Helianthemum songaricum seedlings .The germination rate , germination index and vigor index of Helianthemum songaricum seeds declined with the increasing of PEG solutions .Seed germination was less affected under the 10%and 20%of PEG, and then it was obviously inhibited under 25%, and seeds could hardly germinate under 30%.With the increasing of day numbers without water , the relative water content of leaves declined , the chlorophyll concentration increased firstly and then decreased , and the contents of proline ,soluble protein and MDA increased with obvious variation at the mid-late drought stress stage .

A determination method of ursolic acid in Cynomorium songaricum by reversed phase high performanceliquid chromatography was established. Using a WondaSail C18(4.6nm×150nm,5μm) column. The mobile phase consisted of methanol-0.5%phosphoric acid-acetonitrile(80∶15∶5),flow rate=1mL/min and temperature T=35℃.The detection wave length was at 210nm. With the range of 0.841~8.096μg for ursolic acid had good linearity. The average recovery was 96.54% , RSD=1.8%(n=6). The concentration of ursolic acid in Cynomorium songaricum at 0.466~1.034%, the method is higher accurate,simple and precise.

OBJECTIVE To screen the anti-stress effects of different extracts from Cyrmmorium songaricum and explore its possible anti-stress mechanisms.METHODS The anti-stress effectiveness were investigated from experiments of antifatigue,anti normobaric hypoxia,anti-hyperthermia and anti-hypothermia.The influence to mice organ index of spleen,thymus gland,adrenal glands and testis by aqueous extract were measured.The mice plasma testosterone,estradiol were measured through electrochemiluminescence and cAMP,cGMP were measured through radio-immunity.RESULTS The survival time of mice is longer than positive control group and significantly longer in two Cynomorium songaricum aqueous extracts treated groups of antifatigue and anti-hypothermia than that of blank control group(P 0.05).After treatment of aqueous extract,the contents of cAMP and cGMP were increased,the ratio of cAMP/cGMP was decreased.Meanwbile,the content of testosterone and mice organ index were also decreased while the content of es-
对锁阳熊果酸的乙醇回流提取工艺进行了研究,并采用响应面法优化了提取工艺条件。结果表明:在乙醇体积分数为80%、提取温度78℃、提取时间1.6 h、液料比14∶1(m L∶g)、提取2次的条件下,锁阳熊果酸的提取量为6.43 mg/g,提取率为97.72%。
The ethanol refluxing extraction technology of ursolic acid from Cynomorium songaricum Rupr. was studied. The response surface method was used to optimize the conditions. The result indi?cate that under conditions of ethanol volume fraction 80%, extraction temperature 78℃, extraction time 1?6 h, liquid?solid ratio 14 ∶ 1 ( mL ∶ g) , extraction times 2, the extraction amount of ursolic acid was 6?43 mg/g and the extraction rate was 97?72%.

Objective To optimize the best extraction process of catechin,gallic acid and protocatechuic acid in Cynomorium songar-icum by the orthogonal test. Methods With the contents of catechin,gallic acid and protocatechuic acid in the extracting solution as the observing indexes,the L9 (34 ) orthogonal design was adopted to arrange the experiments for investigating the influences of the solvent type,solvent amount and extracting time on the exacting process. Results The optimum conditions of extraction process were as follows:the medicinal materials were sifted by 40 mesh,adding 5 times amount of water into the materials and ultrasonic extracting for 30 min. Conclusion The optimized extraction process can improve the yield rate of catechin,gallic acid and protocatechuic acid in Cynomorium songaricum,the technology is simple and practical.

Objective To investigate the protective effect and the possible mechanism of Wenshen Shengjing Decoction (WSSJD)(including Cornu Cervi Nippon Parvum , Panax ginseng, Cynomorium songaricum, Cistanche deserticola,Radix Astragali, Epimedium brevicornum, Angelica sinensis.) on cyclophosphamide induced testicular oxidative damage in mice . Methods Eight-week-old male Kunming mice were assigned .Mice in normal control group received intraperitoneally nor-mal saline, and mice in Western medicine group ,WSSJD group and model group were injected intraperitoneally with cyclo-phosphamide [80 mg/(kg? d) ]for five days.Then mice in Western medicine group and WSSJD group were gavaged with clomifene citrate or WSSJD for 30 days consecutively .The epididymal sperm count and sperm viability were recorded , and the structure of seminiferous epithelium was observed .The content of malondialdehyde (MDA) and glutathione (GSH), and activities of glutathione peroxidase ( GSH-Px) and catalase ( CAT)

OBJECTIVE The effect of Cynomorium Songaricum polysaccharide (CSP) on immune function was studied on the aged mice for CSP''s applying for the clinical uses. METHODS Kunming mice were intraperitoneal injected with D-galac-rose to make the aging models, and different dosages of CSP were given by gavage for 56 days. Spleen coefficient and thymus coefficient were determined. MTT method was used to detect the proliferated number of the bone marrow cells. Lymphocyte transformation rate and peritoneal macrophage phagocytic rate in vivo were tested. The activity of telomerase in the testicles of the mile mice were detected by Elisa method. RESULTS Low, middle and high dosages of CSP could cause a significant in-crease of the proliferated number of the bone marrow cells in the aged mice (P<0.01 ).
