本文主要利用三种发根农杆菌A4、15834、R1601诱导苦参产生毛状根,初步为建立苦参的毛状根培养体系提供参考。本实验采用共培养法,考察了不同外植体、菌种、培养时间对苦参毛状根诱导率的影响,并对苦参毛状根中氧化苦参碱进行初步的含量测定。研究结果表明,苦参毛状根的最佳诱导条件是以生长1周的苦参幼芽为外植体,R1601为菌株,添加100μmol/l的乙酰丁香酮,侵染20 min,(25±1)℃暗处诱导,最终测定诱导率最高为27.6%,存活率为45.8%,而苦参毛状根中氧化苦参碱的富集量为1.68mg/g(苦参毛状根干重),未检测到苦参碱。
n this paper,hairy root of Sophora flavescens Ait.were induced by the infection of third Agrobacterium rhizogenes strains 15834,R1601,A4 and an in vitro culture system of the hairy roots was newly established.Hairy roots were induced by co -culture.Effects of explants, Agrobacterium rhizogenes,culture time on growth of hairy root were studied and Determination of Oxymatrine in Sophora flavescens Ait.The research shows that the highest induction frequency was obtained from young shoot with one week which were induced by R1601 for 10mins ,and increase of 100 μmol /L acetosyringone. The transformation frequency was 27.6%,annual survival rate was 45.8%. Hairy roots of Oxymatrine enrichment amount 1.68mg/g ,Total matrine content was not determined.

Objective To explore the effects of matrine and oxymatrine on apoptosis in human hepatocarcinoma SMMC-7721 cells.Methods The MTT assay and double staining of annexin V-fluorescein isothiocyanate(annexin V-FITC)/propidium iodide(propidium iodide, PI)were used to detect proliferation and apoptosis of SMMC-7721 cells, respectively.Results When the concentrations of matrine and oxymatrine were 0.50 mg/ml, 1.00 mg/ml and 2.00 mg/ml, the proliferation inhibition rates in SMMC-7721 cells was gradually increased in a dose- and time-dependent manner,and the inhibition of matrine on proliferation were greater than that of oxymatrine in the same concentration(1.00 mg/ml)(24 h:42.39%±0.04%vs. 21.36%±0.02%;48 h: 51.69%±0.03%vs. 36.16%±0.02%;72 h: 78.98%±0.05%vs. 61.24%±0.13%;allP<0.05). When the concentrations of matrine and oxymatrine were 0.25 mg/ml, 0.50 mg/ml and 1.00 mg/ml, the apoptosis rates of SMMC-7721 cells were significantly increased;and induction of matrine in apoptosis

To investigate the effects of matrine on the JAK - STAT signaling transduction pathways in cervical cancer - derived SiHa cells. Methods:Treated with different concentrations of matrine solution for 24h,48h and 72h,the cytotoxic effect of matrine on SiHa cells was assessed with MTT assay. The protein expression of JAK1, JAK2,STAT3 in SiHa cells were detected by Western blot. Results:Different concentrations of matrine were cytotoxic to SiHa cells in a time - dependent and dose - dependent manner. The protein expression of JAK1,JAK2,STAT3 in SiHa cells with matrine was significantly lower than those in control group(P ﹤ 0. 01). Conclusion:Matrine has a de-pressive role in the proliferation of cervical cancer - derived SiHa cells. Its mechanism is related to down - regulating the protein expression of JAK1,JAK2,STAT3 and inhibiting the signaling transduction pathway of JAK - STAT.

Objective: To optimize the extraction technology for the Sophorae Tonkinensis Radix Et Rhizoma. Methods: Taking the content of the Matrine within the extracts as a study index, we use HPLC to determine its content. Then we compare differences among Water extraction - Alcohol precipitation,60℃Acid water soaking- Alcohol precipitation,pH8-9 Ammonia water extraction Alcohol precipitation and 60% Alcohol Heating reflux method.Result: The content of the Matrine from 60% Alcohol Heating reflux method is the higher.In the range of 6.24μg~399.04μg,the content of the Matrine shows a good linear relationship and the recovery is 100.08%.Conclusion: Using the 60% alcohol Heating reflux method, the content of the Matrine from the Sophorae Tonkinensis Radix Et RhizomaExtracts is the higher, with a highest metastasis rate. The extracting method is reasonable, simple and suitable for industrialized production.
