为了优化东亚飞蝗消化系统双向电泳技术,建立东亚飞蝗消化系统蛋白表达图谱,探讨雌、雄东亚飞蝗消化系统蛋白组分的差异,利用pH值为3~10和pH值为4~7的胶条分别对雌、雄东亚飞蝗消化系统进行双向电泳分析,进行蛋白组学分析.研究结果发现:雌性东亚飞蝗消化系统蛋白种类多于雄性,雌性东亚飞蝗消化系统特异蛋白中偏酸性蛋白数量与偏碱性蛋白相当,而雄性东亚飞蝗消化系统特异蛋白中酸性蛋白多于碱性蛋白.在雌、雄东亚飞蝗消化系统相匹配蛋白中,含量差异达3倍以上的蛋白约占总匹配蛋白的50%.可见,雌、雄东亚飞蝗消化系统蛋白组分存在差异.
Two-dimensional electrophoresis system was optimized and carried out to establish protein expression map of alimentary system from Locusta migratoria manilensis (Meyen) and then the proteomics difference was in-vestigateed between female Locusta migratoria manilensis (Meyen) and male ones. Two-dimensional electrophore-sis was carried in pH 4~7 and pH 3~10 in order to obtain the best condition and to analyze the proteomics differ-ence between female Locusta migratoria manilensis (Meyen) and male ones. To investigate the effect of Bt-transgenic rice on cold resistance of Oriental migratory locust,supercooling point and freezing point of Oriental migratory locust were determined with supercooling point meter after feeding on Bt-transgen-ic rice and non-transgenic rice respectively for three generations in the laboratory conditions.The results showed that the supercooling point of forth instar larvae,fifth instar larvae,female and male adults were -5.89,-7.59,-6.64 and -6.79,respectively,and there were not significant difference with that of control.

The activity of ligarine extracts from Stellera chamaej asme L .against Locusta migratoria manilensis(Meyen) was tested and compared by bioassay methods . In order to explore the functional genes of Metarhizium anisopliae related to virulence, a T-DNA containing benomyl resistance gene benA3 was transferred into M. anisopliae strain IMI330189 to construct mutant library by using Agrobacterium-mediated transformation method. The toxicity of some mutants to the third instar larvae of Locusta migratoria was tested at a concentration of 108 spores·g?1 with artificial bait method. The toxicity of mutant 189-1 was slightly shorter with the LT50 3.5 d compared to the wild strain IMI330189 with LT50 4.3 d. The mutants 189-4, 189-5, 189-6, 189-10, 189-11, 189-13 and 189-15 almost lost their toxicity to locust, the corrected mortality rates were less than 50%. By constructing mutant library of M. anisopliae strains IMI330189, we obtained some mutants with virulence lack. The study provided a tool to isolate virulent genes and explore the pathogenic mechanism of M. anisopliae.

We reared oriental migratory locusts, Locusta migratoria manilensis (Meyen) in the laboratory from egg stage until egg-laying female adults, at six different degree-days (544.3, 662.2, 678.8, 760.2, 840.0 and 843.5 heat units), to reveal the influence of degree-days (DD) on diapause rate of overwintering locust eggs. Results showed that the diapause rate of eggs at the six treatments was 73.1%, 58.6%, 48.6%, 12.1%, 0 and 0, respectively. This indicated there were only parts of overwintering locust eggs could enter diapause during winter. The relationship between diapause rate of eggs (D) and DD of pre-overwintering period (Y) was D=?0.719Y+82.75, R2=0.774. When Y≥115.1 DD, all overwintering locust eggs would hatch. We further studied the sensitive temperature on diapause rate of eggs at the six treatments, results showed that DD based on temperature over than 27 ℃ (E) had the significant effect on diapause rate of eggs (D), the relationship was D=?0.058E+0.732, R2=0.967.
