In order to ascertain theoretical basis of fatty acid metabolism mechanism in Vernicia fordii, taking seeds of a ifne variety‘Grape Tong’ as materials, its transcriptome database was established, the transcriptome and expression spectrum database were comprehensively analyzed, total 83 non-redundant gene sequences were found, which involved in fatty acid metabolism in V. fordii and 10 main enzyme genes. Taking KEGG database as a reference, the fatty acid metabolism pathways in V. fordii were analyzed, and its pathway graph was drawn, which revealed the basic rules of key enzyme genes regulation and catalytic fatty acid metabolism in fatty acid metabolism pathways in V. fordii.

In order to make full use of Vernicia fordii flower resources and to enrich landscape plants, selecting 53 V. fordii lines in State Genetic Resources Preservation Library of Xiangxi Autonomous Prefecture as research objects, ifve ornamental indexes were investigated in the forth year after afforestation. Ornamental values of different lines of V. fordii flowers were comprehensively evaluated by using the method of fuzzy mathematics. The results show that the four strains of‘CSUFT No.19’,‘CSUFT No.10’,‘CSUFT No.13’ and‘CSUFT No.16’ have higher ornamental values.

[Objective]The full-length sequence of acid phosphatase(ACPase)gene in tung tree was cloned to provide ref-erences for further studies on the functions of ACPase. [Method]The primers were designed according to the full-length se-quence of acid phosphatase gene in transcriptome of tung tree. ACPase gene from tung tree seeds was cloned by RT-PCR. Bioinformatic analysis was used to predict cDNA sequence and its deduced nucleotide sequence similarity, physicochemical properties, hydrophobicity, transmembrane structure, secondary structure, and tertiary structure of ACPase. Multiple se-quences alignment and phylogenetic trees were also constructed. [Result]ACPase in tung tree was cloned. The largest open reading frame of ACPase had 1152 bp in length and encoded a polypeptide of 383 amino acid residues. By using the real-time fluorescent quantitative PCR technique, the identification of expression of DGAT1 in different vareties and seed developmental stages of Vernicia fordii was carried out. The results indicate that DGAT1 relative expression quantity was no distinct differences in the six V. fordii matured seeds,and this expression was not obvious correlation with oil content;DGAT1 transcripts was relatively low from June to July, with higher levels on August, September and October, the relative expression quality were up-regulated in different stages of‘ZNL-F52’ seeds and this was consistent with V. fordii seed kernel oil formation law, which suggests that this gene may have a close relationship with the regulation of the synthesis of Tung oil.

In order to reveal sequence features and structural function of accA gene in Vernicia fordii, and to lay a foundation for further research, development and utilization, taking nearly ripe seeds in V. fordii as materials, the speciifc primers were designed according to the results of transcriptome sequencing analysis of V. fordii, and full-length cDNA sequences of accA gene were cloned by RT-PCR technique. The results show that full-length cDNA sequence of accA gene is 2 313bp, encoding 770 amino acids. Iso electric point (pI) of accA protein is 8.48, the relative molecular mass is 85 902.7 Da, and the stability coefifcient is 37.33. By taking PUPUTUNG tree seeds as the tested materials, according to the known part’s (enoyl-CoA reductase, ECR) sequence and by using RACE technology, PUPUTUNG tree seed’s ECR gene was cloned and the gene’s full-length cDNA was obtained, named VfECR.
