龙胆科龙胆属滇龙胆( Gentiana rigescens)与獐牙菜属青叶胆( Swertia mileensis)为我国特有种。采用高效液相色谱法测定其中马钱酸、獐牙菜苦苷、龙胆苦苷和当药含量,结合多变量分析研究环烯醚萜类成分含量及其构成比例在龙胆科植物种内和种间的差异。研究结果显示,4种环烯醚萜类成分的计量特征呈现种间差异,依据环烯醚萜类成分含量及其构成比例,所有滇龙胆样品可分为4类。马钱酸含量与纬度呈极显著负相关( R=-0?348, P0?05)。
Gentiana rigescens and Swertia mileensis belonged to Gentianaceae are endemic plants in China. Four iri ̄doids ( loganic acid,swertiamarin, gentiopicroside and sweroside) in G?rigescens and S?mileensis were determined by HPLC ̄DAD. Their chemometrics characterizations were analyzed with multiple statistical analyses. The results showed that there were differences of chemometrics characterization of the iridoids between the two species. All the samples of G?rigescens could be classified into four groups based on contents and percentage contents of the four iri ̄doids. Content of loganic acid had significant negative correlation with longitude ( R=-0?348, P 0?05) in the study.

利用红外光谱三级鉴定方法对藏药川西獐牙菜及其不同溶液提取物进行分析研究。选择川西獐牙菜中含量相对较高的药用成分龙胆苦苷作为不同溶液提取物的对照成分进行红外光谱和二阶导数光谱的分析;同时对不同浓度乙醇提取物进行二维相关光谱分析。结果显示,龙胆苦苷红外光谱中1 611和1 075cm-1处的两个主要特征峰强度的变化趋势,和川西獐牙菜不同溶剂提取物中龙胆苦苷含量变化趋势一致;提取溶剂极性相近时,提取物的红外光谱图相似;随着提取溶液极性的逐渐降低,提取物的红外谱图中2853,1 733,1 464,1 277和1 161cm-1等处吸收峰强度逐渐增强,提取物中酯类化合物含量逐渐增加,萜类化合物的提取率逐渐增加;红外光谱图极为相近的不同浓度乙醇提取物在二维红外光谱图中有很明显的差异。龙胆苦苷特征峰强度的变化趋势和不同提取物中龙胆苦苷含量变化趋势的一致性说明红外光谱分析结果能体现分析对象中所含有成分的含量变化;川西獐牙菜原药材,水提物和不同浓度乙醇提取物谱图的相似性和相似度的变化规律,说明传统用药水煎液和低浓度醇提物的科学性;红外光谱技术可成为川西獐牙菜不同提取物后期药理活性成分宏观控制,和多以复方入药的藏药研究中准确、快速、有效的分析方法。
The objective of the present study is to research the herb of Swertia mussotii Franch and its different extracts by tri-step infrared spectroscopy .The main constitute of Swertia mussotii Franch-gentiamarin ,which is also the higher content consti-tute ,was selected as the control components to analyze the infrared spectroscopy and second derivative infrared spectroscopy of different extracts of Swertia mussotii Franch ,at the same time ,the different concentration of ethanol extracts were also analyzed by two-dimensional correlation spectroscopy(2D-COS) .The results indicated that the intensity of 1 611 and 1 075 cm -1 of gen-tiamarin ,which are its two main absorptions in the infrared spectra ,has the positive correlation with the content change in dif-ferent extracts .The infrared spectroscopy of extracts are similar if the polarity of extract solvents is close;with the decreases in solution polarity ,the intensity of 2 853 ,1 733 ,1 464 ,1 277 and 1 161 cm-1 in infrared s

本实验建立了HPLC波长切换法同时测定云南丽江粗茎秦艽中马钱酸、獐牙菜苦苷、龙胆苦苷獐牙菜苷、异荭草、异牡荆6种成分的含量,并对各成分含量间的相关性进行分析,为粗茎秦艽药材质量的全面评价提供了科学依据。采用Sepax Gp-C18(150 mm×4.6 mm,5μm)色谱柱,甲醇-0.1%磷酸为流动相体系,梯度洗脱,流速1.0 mL·min-1,柱温25℃,检测波长235 nm(0~51 min)、270 nm(51~57 min)、243 nm(57~65 min)、270 nm(65~80 min)。马钱酸、獐牙菜苦苷、龙胆苦苷獐牙菜苷、异荭草、异牡荆的量分别在0.01000~10.0000、0.00216~2.1600、0.0480~48.0000、0.0010~1.0000、0.00034~0.3400、0.00026~0.2600μg范围内与色谱峰峰面积呈良好的线性关系,平均回收率为98.52%~99.31%,RSD均小于3.5%(n=6);对各成分含量间相关性分析发现环稀醚萜类与黄酮类组分含量间无相关性,四种环烯醚萜类成分的含量之间存在着显著相关性,两种黄酮类成分的含量之间也存在相关性。该方法简便、快速、准确,重现性好,更符合中药材复杂体系的多指标质量控制发展趋势,为粗茎秦艽药材的质量控制找到了简便易行的方法。
To establish an HPLC with switching wavelength method for determination of six components( loganic acid,swertiamarin, gentiopicroside,sweroside,isoorientin,isovitexin)in Gentiana crassicaulis. Sepax Gp-C18(150 mm × 4. 6 mm,5 μm)column was used,the mobile phase was methanol-0. 1%phosphoric acid solution with gradient elution at a flow rate of 1. 0 mL·min-1 . The col-umn temperature was set at 25℃,detection wavelength was 235 nm(0~51 min)、270 nm(51~57 min)、243 nm(57~65 min)、270 nm(65~80 min). The six components(loganic acid,swertiamarin,gentiopicroside,sweroside,isoorientin,isovitexin)content in the ranges of 0. 01000 ~10. 0000,0. 00216 ~2. 1600,0. 048 ~48. 0000,0. 0010 ~1. 0000,0. 00034 ~0. 3400 and 0. 00026 ~0. 2600 μg respectively,had a good linearity. The average recoveries were 98. 52% ~99. 31%,the RSD was less than 3. 5%. 目的建立高效液相色谱方法测定肝泰舒胶囊中獐牙菜苦苷含量的方法。方法采用Waters Symmetry-C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.02%磷酸为流动相进行梯度洗脱,流速为1.0 mL/min,检测波长为250 nm,柱温为室温。结果獐牙菜苦苷在0.373 556~2.988 448μg范围内呈良好的线性关系(r2=1.000 0),平均回收率为106.81%,RSD=3.7%。结论该方法快捷、准确、简便、重复性好,可用于肝泰舒胶囊的质量控制。
Objective To establish an HPLC method for determining the content of swertiamarin in Gantaishu capsules. Methods Using a Waters Symmetry C18 column (4.6 mm×250 mm, 5μm), the mobile phase was acetonitrile-0.02%phosphoric acid, with flow rate of 1.0 mL/min. The detection wavelength was at 250 nm, the column temperature was room temperature. Results The content of swertiamarin showed a good linear relationship in the range of 0.373 556-2.988 448 μg, r 2=1.000 0. The average recovery was 106.81%, and RSD was 3.7%. Conclusion This method is fast, accurate and simple with good reproducibility, which can be applied to the quality control of Gantaishu capsules.

This paper aims to establish a new comprehensive method to evaluation the quality of gentiana macrophylla which collect-ed from 17 different regions in LIjiang. We established HPLC fingerprint and tested 9 quality indexs of the samples. HPLC finger-print was established with mobile phase consisting of methanol and 0. 1%phosphoric acid;The loganic acid,swertiamarin and genti-opicroside were detected by HPLC method;The ethanol-soluble extractives,total ash and acid insoluble ash were determined accord-ing to the pharmacopoeia method;Spectrophotometric method was used to determine polysaccharide content.
