为明确威宁中华蟾蜍的分子分类学地位,对采自贵州省威宁县的蟾蜍标本(标本号:LPSWN01A)进行线粒体12S rRNA 基因扩增和双重单系法分析。结果表明:该标本的序列大小为906 bp (GenBank 登录号:KJ803016),基于数据集 A 构建的系统发育树中,威宁中华蟾蜍与7条中华蟾蜍同源序列聚为一个单系(中华蟾蜍支系),威宁中华蟾蜍以较低的最大似然率独立为一个支系;基于数据集 B 构建的系统发育树中,威宁中华蟾蜍与2个中华蟾蜍华西亚种种群聚为一支,因此推断威宁产蟾蜍属于中华蟾蜍华西亚种(Bufo gararizans andrewsi )。另外,威宁中华蟾蜍与云南怒江种群的遗传距离最近,为0.0093,说明威宁中华蟾蜍属于中华蟾蜍华西亚种,与之前的传统分类文献记载一致。
The mitochondria 12S rRNA gene of Weining B.gargarizans sample (LPSWN01A)was amplified and analyzed by dual monophyletic method to definite molecular taxonomic status of B. gargarizans.The sequence length of Weining B.gargarizans is 906bp (GenBank accession number:KJ803016).The sequence of Weining B.gargarizans and seven homologous sequences of B.gargarizans form a monosystem in the phyogenetic tree built by data set A. Weining B.gargarizans forms an independent clade with lower maximum likelihood. Objective To investigate the effects of bufalin and cisplatin on proliferation and apoptosis of tongue squa -mous cell line Tca8113.Methods Tongue squamous cell line Tca8113 was cultured and the logarithmic phase cells were divided into control, bufalin(10,20,40,80,160 nmol/L), cisplatin(2.5,5,10,20,40 μg/mL) and combination groups. MTT assay was used to examine the inhibition rate of cell growth when cell were treated at various concentrations of bufalin and cisplatin alone or in combination and calculate inhibiting concentration of 50%cell growth ( IC50 ) .The corresponding concentrations of three drug groups were selected based on IC 50 of Bufalin and cisplatin at 48 h.Apoptosis was evaluated by flow cytometry.The protion expression of Bax ,Bcl-2,Caspase-3,P-ERK and Erk were examined by Western blot method . By applying biological functional test systems,this paper aims to study the influences of the different concentrations of Toad Venom on the heart rate and electrocardiog ram(ECG)of Bufo gargarizans. Results show that,as Toad Venom concentrations increase,the heart rate of the bufo gargarizans speeds up;the peak voltage of R wave and the time of the Q-T interval in the ECG decreases;while the time of the P-R interval and QRS dura-tion increases. These values after administration of 1 ~ 1. 5 h reach the maximum,and each value decreased at 2 h. Thus,it can be concluded the toad venom has an effect on the elcetrocardia action of bufo gargarizans.

Bufadienolide is a kind of natural steroids with significant anti-cancer activities. Hitherto, the number of structural modification on the bufadienolides has reached more than 100 . The purifica-tion of sulfated,a trace cytotoxic component bufadienolides from toad skin is a tough work due to its low content and unstable structures. However,there is no report on synthesis of bufadienolides sul-fonation though they have promising cytotoxic activities. In this paper,the arenobufagin was firstly taken as an example,to develop an effective method for the synthesis of arenobufagin-3-O-sulfonate. In order to enrich the arenobufagin-3-O-sulfonate quickly and mildly,a reversed phase/hydrophilic solid-phase extraction method based on our homemade hydrophilic Click TE -Cys was developed. The structures of arenobufagin-3-O-sulfonate and arenobufagin were both identified by MS,1 H NMR and 13 C NMR spectra. 筛选出适于监测无束缚蟾蜍动态心电的导联方式,并分析其实用性。方法:蟾蜍在麻醉状态下皮下包埋引导电极后,通过BL-420S生物机能实验系统监测了5种导联的动态心电,比较并筛选出能够较好地显示心电波形的导联。利用该导联监测了蟾蜍从人工冬眠中复苏6 h内的动态心电和连续5 d内的心率和心率变异性(heart rate variability,HRV)的日间稳定性,以及冻融处理前后的心率和HRV,以分析其实用性。结果:5种备选导联中有2种导联能较好显示心电波形。与人工冬眠复苏6 h比较,复苏1 h之内的心率降低,窦性心率RR间期标准差(the standard deviation of normal R-R intervals,SDNN)升高,二者均有显著差异(P0.05或P0.01);2h到5 h之间的心率和SDNN无显著变化,提示心电在复苏2 h后恢复稳定。冬眠第4天和第5天的2 h和4 h心率较第1天显著减慢(P0.05或P0.01),而SDNN较第1天显著升高(P0.05或P0.01),提示留置电极的蟾蜍心电在3 d内不会有显著变化。与冷冻前比较,解冻1 h和12 h的心率显著加快(P0.05),SDNN显著变小(P0.05),表明冻融后蟾蜍心功能下降。结论:皮下植入电极的方法可有效监测蟾蜍动态心电。
[ ABSTRACT] AIM:To screen out a suitable lead for monitoring the ambulatory electrocardiogram ( ECG) in un-restrained toad, and to investigate its practicability.METHODS:After subcutaneously implanting the electrodes in toads under anaesthesia, the ambulatory ECG of 5 leads were monitored with BL-420S data acquisition and analysis system, and the leads which could well express the waveform in ECG were screened out.The recovery process of the toads from the arti-ficial hibernation within 6 h, the day-to-day stability of the heart rate ( HR) and the heart rate variability ( HRV) in 5 suc-cessive days of hibernation, and the HR and HRV after freeze-thawing process were monitored to determine its practicabili-ty.RESULTS:Two out of 5 leads showed better ECG waveforms.Compared with 6 h post hibernation, lowered HR at 0 h and 1 h was observed, and the standard deviation of normal R-R intervals ( SDNN) was significantly increased ( P<0.05 or P<0.01), but the HR and SDNN from 2 to 5 h

The observations and analysis of Bufo melanostictus karyotype, Ag-NORs were based on the chromosome sample from the bone marrow cells. The results showed that:the number of chromosome was 2n=22 (20M+2SM), NF=44, with 11 pairs of chromosomes consisting of 6 pairs of megachromosome and 5 pairs of minisome. No heteromorphic chromosome was found correlating with sex differentiation. No region of the Ag-NORs chromosome was found. Compared with the reported research about B. melanostictus karyotype from Guangzhou Guangdong, Fuzhou Fujian, Nanning Guangxi, Wenzhou Zhejiang, Tengchong Yunnan, Taiwan and other different regions, it showed that there were some differences about B. melanostictus karyotype from different regions in the SM number and the order, secondary constriction and satellite, Ag-NORs areas and so on.

为了解上海市市售蟾蜍寄生蠕虫的感染情况,对60只中华大蟾蜍进行剖检,按脏器检查、收集寄生虫虫体,统计寄生虫的感染率与感染强度。结果显示,蟾蜍寄生虫感染率为100%,平均感染强度为28.23条。不同种类寄生虫的感染情况明显不同,线虫、吸虫、绦虫和棘头虫的感染率分别为93.33%、73.33%、8.33%和15.00%,感染强度分别为17.52、14.48、10.40和2.67条。不同脏器寄生虫的感染情况不同,肠道和肺脏的寄生虫感染率最高,分别为100%和88.33%;肠道、肺脏、肌肉寄生虫的感染强度分别为15.63、12.28和11.33条,明显高于其他器官。不同种类寄生虫的寄生虫部位明显不同,线虫主要来源于肺部和肠道,吸虫和棘头虫主要来源于肠道,绦虫主要来源于肌肉。通过对检出虫体的形态进行初步观察,吸虫、线虫、绦虫和棘头虫种类分别约有3、4、1和1种,但具体种类尚需进一步鉴定。本调查结果为掌握上海市市售蟾蜍寄生虫感染状况和做好寄生虫病防控提供了基础数据。
To investigate infection status of parasites in toads at Shanghai farmer’s markets, organs and tissues (subcutaneous, muscles, hearts, lungs, livers, stomachs, intestines) from 60 toads (Bufogargarizans) were collected and examined under microscopy. All 60 toads were confirmed to harbour parasites and the average infection intensity (number of parasites per toad) was 28.23. The infection rates/intensities of nematodes, trematodes, tapeworms and acanthocephalan were 93.33%/17.52, 73.33%/14.48, 8.33%/10.40, and 15.00%/2.67, respectively. The infection status significantly varied in organs and tissues. The highest infection rates were found in intestines (100%) and lungs (88.33%). The infection intensities of intestines, lungs and muscles were 15.63, 12.28 and 11.33, respectively. The parasite species differed among organs and tissues. The nematodes were mainly found in lungs and intestines, trematodes and acanthocephalan in intestines and tapeworms in muscles. Species numbers
