A browser-based EPUB3.0 reader application is developed with HTML 5,CSS3 and JavaScript technolo-gies.The algorithm for parsing OPF documents and content documents , and the theory of EPUB Canonical Frag-ment Identifier(CFI) are discussed in detail.This EPUB3.0 reader is able to support more media types and seman-tics than other similar applications .

背景:间充质干细胞作为种子细胞广泛应用于组织工程和临床治疗,而安全、高效的分选方法是间充质干细胞建系的前提。 目的:综述近年来间充质干细胞分选方法的研究进展,比较各方法的优缺点,为安全、高效分离间充质干细胞提供理论依据。 方法:第一作者检索 CNKI 数据库(http://epub.cnki.net/)和 PubMed 数据库(http://www.ncbi.nlm.nih.gov/pubmed/)的相关文献。中文检索词为“间充质干细胞、分离方法”;英文检索词为“mesenchymal stem cel s (mscs),isolation methods”。查阅1965至2014年来间充质干细胞分离方法的相关文献,包括综述、临床研究和基础研究,最终纳入52篇文献。 结果与结论:①全骨髓贴壁法可快速获得大量间充质干细胞,但需要进一步纯化。②梯度密度离心法选择密度为1.073 g/mL的介质在原代能得到相对较纯的间充质干细胞。③组织消化法适用于脂肪、脐带等组织进行消化分离,选用胶原酶Ⅱ消化效果更佳,但是均存在操作技术要求高的缺陷。④免疫磁珠法适用于研究表达特定细胞表面标记物的某一个间充质干细胞亚群的生物学特性。⑤各种分离方法相结合也是一种理想选择。⑥对于近年来出现的分离新方法,由于案例有限,其可靠性有待进一步研究。
BACKGROUND:Mesenchymal stem cel s as potential seeded cel s have been widely used in tissue engineering and clinic therapy;thus, the precise, safe, effective isolation of mesenchymal stem cel s is the particular important premise to build culture system. OBJECTIVE:To review the methods of isolating mesenchymal stem cel s and to compare the merit and demerit of different methods, thereby providing theoretical basis for safe and high-effective isolation of mesenchymal stem cel s. METHODS:A computer-based online research of CNKI and PubMed databases was performed to col ect articles, which included reviews, clinical trials and experiments, published between 1965 and 2014 with the key words of“mesenchymal stem cel s (MSCs), isolation methods”in Chinese and English. A total of 52 articles were included according inclusion and exclusion criteria RESULTS AND CONCLUSION:(1) The whole bone marrow culture method can derive a mass of mesenchymal stem cel s, which need to be purified.
