目的:探讨 OK432优化的人脐静脉内皮细胞( HU-VECs)疫苗对小鼠EAC乳腺癌的生长抑制作用。方法体外培养HUVECs,与佐剂OK432混合,制备HUVECs-OK432疫苗,以小鼠 EAC 乳腺癌皮下移植瘤模型考查 HUVECs-OK432疫苗的抗肿瘤效应,并通过ELISA、脾细胞增殖及细胞毒性T淋巴细胞( CTL)杀伤实验检测疫苗免疫后体液及细胞免疫应答水平。结果在预防性免疫中, HUVECs-OK432疫苗可以明显抑制EAC乳腺癌生长;HUVECs-OK432疫苗诱导小鼠产生了高滴度的特异性 HUVEC 抗体;HU-VECs-OK432疫苗能有效刺激免疫小鼠脾淋巴细胞的增殖;HUVECs-OK432组小鼠脾细胞诱导产生了明显的靶向HU-VEC细胞的CTL杀伤作用。结论 HUVECs-OK432疫苗可以有效诱导机体产生靶向HUVEC的特异性体液及细胞免疫应答,从而有效抑制了小鼠EAC乳腺癌的生长。
Aim To investigate the anti-EAC breast cancer effects of viable human umbilical vein endothe-lial cells ( HUVECs) vaccine. Methods Subconflu-ent HUVECs were mixed with OK432 to prepare HU-VECs-OK432 vaccine, and the anti-tumor efficacy of HUVECs-OK432 was investigated using a subcutaneous tumor model of EAC breast cancer. ELISA, splenic lymphocyte proliferation assay and cytotoxic T lympho-cytes ( CTL ) killing assay were adopted to detect the humoral and cellular immune responses after HUVECs-OK432 immunization. Results HUVECs-OK432 im-munization significantly inhibited the growth of EAC tumor in mice in the prophylactic procedures. High ti-ter of anti-HUVEC antibody was elicited by HUVECs-OK432 immunization. The proliferation activity of splenocytes from mice immunized with HUVECs-OK432 was significantly increased. 目的利用DBA/2小鼠移植舌癌荷瘤模型,探究OK-432肿瘤细胞疫苗对舌癌的抑制作用。方法以戊二醛为交联剂,使KLN-205细胞与OK-432产生交联,制备OK-432肿瘤细胞疫苗;将DBA/2小鼠随机分为4组,每组20只,分别右侧腹部皮下注射0.9%氯化钠注射液、OK-432、灭活的KLN-205细胞、OK-432疫苗,接种KLN-205肿瘤细胞后观察对比舌癌瘤体大小,并用统计学分析其结果。结果肿瘤大小除KLN-205组与对照组间差异没有统计学意义(P=0.094)外,其余各组间差异均有统计学意义,其中OK-432疫苗组肿瘤体积均小于其他3组。结论 OK-432肿瘤细胞疫苗能抑制小鼠舌癌的生长,具有一定的抗肿瘤效应。
Objective To investigate the inhibitory effects of OK-432 tumor cell vaccine on tongue cancer growth in DBA/2 mice by using the tongue cancer xenograft models. Methods The OK-432 tumor cell vaccine was prepared by inducing conjugation of OK-432 to squamous cell carcinoma (KLN-205) with 0.2% glutaraldehyde (GA). The DBA/2 mice were randomly allocated to receive subcutaneous injection of saline ( n=20), OK-432 (n=20), inacti-vated KLN-205 cells (n=20) and OK-432 tumor cell vaccine (n=20) on the right abdomen. This allowed assessment of the tumor size before and after KLN-205 cell inoculation. Results The tumor size was significantly different among the four groups except for OK-432 vaccine group compared with control group (P=0.094). Inoculation with OK-432 vaccine resulted in marked reduction in the tumor size compared with the remaining three groups. Conclusion OK-432 tumor cell vaccine can inhibit the growth of tongue cancer in mice, suggesting the potential anti-tumor effects.

目的 探讨经尿道膀胱肿瘤电切术联合沙培林膀胱黏膜下注射治疗浅表性膀胱肿瘤的疗效.方法 将85例膀胱肿瘤患者分为两组(A组和B组),A组42例,B组43例,两组均行经尿道膀胱肿瘤电切术,A组另用5KE沙培林溶于50ml0.9% Nacl溶液行膀胱黏膜下注射.两组术后均行沙培林膀胱灌注,膀胱内灌注保留2h,每周1次,连续8周,以后每月灌一次,连续10个月.根据随访结果对两种方法的疗效进行评价.结果 A组平均随访24个月,B组平均随访26个月.A组随访期内复发4例(9.52%),B组随访期内复发16例(37.2%),A组复发率与B组比较,差异有统计学意义(P<0.05).结论 经尿道电切联合沙培林黏膜下注射可减少膀胱肿瘤复发,操作简单,安全有效.
Objectives To investigate the efficacy by studying electrovaporization of bladder cancer and transurethral injeetan of OK-432.Methods 42 case of bladder cancer received the OK-432 solution injected to the pedicle of bladder tumor after operation,and 43 cases only received operation,do not give the OK-432 solution injected to the pedicle of bladder tumor.Both groups received OK-432 routine treatment,the effects Were evaluated according to recurrence ratio.Results Injection group had a mean follow-up of 24 months,control group had mean follow-up of 26 months,The recurrence rate of injection group was 9.52% (4/42),and that of the control group was 37.2 (16/43).There is a very significant difference between the two groups(P <0.05).Conclusions Operation combined with OK-432 injection are simple and safe for the treatment of superficial bladder cancer,can increase the efficacy,and reduce recurren rate.
